Tenoxicam, a thienothiazine derivative of Oxycam, is a medicine from the NSAID group. In addition to its anti-inflammatory, analgesic and antipyretic effects, the drug also inhibits platelet aggregation. Tenoxicam exerts its anti-inflammatory effect by inhibiting the activity of COX isoenzymes involved in the metabolism of arachidonic acid, thereby inhibiting PG synthesis. Tenoxicam has no effect on lipoxygenase activity. In addition, tenoxicam inhibits several leukocyte functions, including phagocytosis and histamine release, and reduces active radicals at the site of inflammation.
The drug is rapidly absorbed from the gastrointestinal tract in unchanged form. CMax is achieved 2 hours after taking the drug. If the drug is taken after a meal or with antacids, the rate of absorption is reduced, but not the extent of absorption. The median of T1/2 is 70 hours. Tenoxicam is completely absorbed; its bioavailability is 100 %. In the blood, 99 % of the active ingredient is bound to proteins.
The drug penetrates well into the synovial fluid, has a low systemic clearance and a long T1/2 so that tenoxicam can be taken once a day.
2/3 of the administered dose is excreted in the urine and 1/3 in the faeces. No accumulation of tenoxicam is observed during long-term use; In this case, the active ingredient content in the serum is 10-15 µg/ml.
